Transforming growth factor beta-1 (TGF-β1) is a common component of human pluripotency maintenance media. However, as TGF-β1 is a highly complex protein, it is difficult to manufacture in microbial expression systems.

Qkine TGF-β1 PLUS™ 14 day stability
- TGF-β1 PLUS™ stored at stored at room temperature (EC50 66.20 pg/ml) didn’t lose bioactivity compared to -80 °C storage (EC50 72.59 pg/ml).
- No loss of activity was seen with TGF-β1 PLUS™ stored at room temperature with multiple freeze thaws (EC50 53.27 pg/ml) compared to -80°C (control, EC50 57.29 pg/ml).
- SDS-PAGE showed no degradation of TGF-β1 PLUS™ was visible after 14 days under these conditions.
TGF-β1 PLUS™ remained stable and bioactive for 14 days at room temperature and with multiple freeze-thaws. Bioactivity was determined using a TGF-β1-responsive (CAGA) firefly luciferase reporter in transiently transfected HEK293T cells. Cells were treated in triplicate for 6 hours with a serial dilution of TGF-β1PLUS™ (Qk010). (A) Stored at room temperature (RT) or -80 °C (control). (B) Stored at room temperature with 4 freeze-thaw cycles (RT/FT) or -80°C (control). Firefly luciferase activity was measured and normalized. (C) 3 µg was resolved on a 15% w/v SDS-PAGE gel under non-reduced conditions and stained with Coomassie Brilliant Blue R250. Data from Qk010 lot #204762.
