Reconstituted animal origin-free GDNF has excellent stability under standard storage conditions
Glial cell line-derived neurotrophic factor (GDNF) is a member of neurotrophin family. GNDF is used to maintain neurons and cortical organoids and to differentiate dopaminergic neurons from human pluripotent stem cell-derived neural progenitors. GDNF also facilitates the differentiation of neural progenitors to astrocytes.
Qkine GDNF (Qk051) is animal origin-free, carrier protein-free and tag-free to ensure high and consistent bioactivity.
Qkine GDNF one month stability
- GDNF diluted to 50 µg/ml and stored in PBS + 0.1% BSA at -20 °C for one month had an EC50 of 18.7 ng/ml, equivalent to freshly reconstituted protein stored lyophilized at -80 °C, which had an EC50 of 18.5 ng/ml.
- When the 50 µg/ml GDNF in PBS + 0.1% BSA was stored at 4 °C for one month it had an EC50 of 16.5 ng/ml, equivalent to the freshly reconstituted protein, which had an EC50 of 24.1 ng/ml.
- When diluted further to 1 µg/ml in cell culture medium and stored at 4 °C for one month GDNF retained an EC50 of 17.6 ng/ml, equivalent to freshly reconstituted protein, which had an EC50 of 19.1 ng/ml.
GDNF remained bioactive under all storage conditions. Bioactivity was determined using an SH-SY5Y cell proliferation assay; cells were incubated with different concentrations of GDNF (Qk051) in the presence of retinoic acid and recombinant GFR α1 for 3 days before viable cell measurement using an MTS assay. (A) Bioactivity of control GDNF (freshly reconstituted and diluted from protein stored lyophilized at -80 °C) compared with protein stored in PBS + 0.1% BSA at -20 °C for one month. (B) Bioactivity of a control protein compared with protein stored in PBS + 0.1% BSA at 4 °C for one month. (C) Bioactivity of control protein compared with protein stored in cell culture medium at 4 °C for one month.
