Qkine bone morphogenetic protein 4 (BMP-4) is stable and bioactive up to 7 days in cell culture conditions
Human bone morphogenetic protein 4 (BMP-4) is a key regulator of embryogenesis and supports the differentiation of induced pluripotent stem cells (iPSC) and embryonic stem cells (ECS). BMP-4 plays a role in bone and cartilage formation, tissue repair, and organ and neuronal development.
BMP-4 is part of the TGF-β superfamily and has a complex biochemical structure, expertise is required to manufacture animal origin-free bioactive BMP-4.
Qkine BMP-4 stability
- After 7 days Qkine BMP-4 (Qk038) bioactivity was equivalent to fresh protein, with an EC50 at day 0 of 51.2 pM, day 2 EC50 53.6 pM and day 7 EC50 40 pM.
- No degradation of protein could be seen in an SDS-PAGE gel after 2, 5 or 7 days.
Stability and bioactivity of Qkine BMP-4. SDS-PAGE of recombinant BMP-4 (A), protein was not degraded when incubated in HEK293T conditioned media for 7 days. Bioactivity was determined using the BMP-4-responsive firefly luciferase reporter assay (B). Transfected HEK293T cells were treated in triplicate for 6 hours with a serial dilution of BMP-4 which had been pre-incubated in conditioned media for 2 or 7 days. Firefly luciferase activity was measured and normalized to the control Renilla luciferase activity.
